Caroline Goujon’s team focuses on uncovering novel human genes that regulate RNA virus replication, with a particular emphasis on the intrinsic and innate defense mechanisms of host cells. By combining reverse and forward genetic approaches, including genome-wide CRISPR screens, they have identified novel genes that control the replication of major human pathogens such as HIV-1, influenza A virus, and SARS-CoV-2, which are now under detailed characterization. A key ambition of the team is to extend these screening strategies to primary cellular models, thereby potentially bringing discoveries closer to physiological contexts. In parallel, they investigate the mechanisms of well-established antiviral restriction factors, such as the high molecular weight GTPase MX1, a potent inhibitor acting against a broad spectrum of RNA and DNA viruses.​
